RESUMO O objetivo deste trabalho foi avaliar a atividade antimicrobiana dos extratos vegetais de Ocotea silvestris Vattimo-Gil e Ocotea diospyrifolia (Meisn.) Mez frente a 10 sorotipos de Salmonella enterica de origem avícola de maior incidência na região Oeste do Paraná, bem como realizar a triagem fitoquímica dos compostos de Ocotea silvestris Vattimo-Gil e Ocotea diospyrifolia (Meisn.). Após a obtenção do extrato aquoso e dos extratos orgânicos (álcool etílico, acetato de etila e hexano) foram determinadas a concentração inibitória mínima (MIC) e a concentração bactericida mínima (MBC) por meio da técnica de microdiluição em caldo. A prospecção fitoquímica foi realizada de acordo com metodologia qualitativa de precipitação e colorimetria. Os extratos apresentaram atividade inibitória e bactericida em diferentes concentrações (200 a 25 mg.mL-1) frente aos sorotipos de Salmonella, variando de acordo com o solvente extrator. O extrato acetato de etila apresentou os melhores resultados, seguido pelo extrato alcoólico, aquoso e hexânico. Ressalta-se que estudos de atividade antimicrobiana com extrato vegetais dentro do gênero Ocotea são escassos, contudo, a família Lauraceae apresenta significativas atividades biológicas, principalmente relacionadas aos seus óleos essenciais. A prospecção fitoquímica detectou a presença de esteróides e alcalóides em ambas as espécies de Ocotea, podendo justificar o potencial antimicrobiano encontrado para os sorotipos de Salmonella. Conclui-se que estas plantas representam uma alternativa para o controle de Salmonella no setor avícola, refletindo uma nova perspectiva para estudos com produtos naturais.

SUMMARY The aim of this work was to evaluate the antimicrobial activity of plant extracts Ocotea silvestris Vattimo-Gil and Ocotea diospyrifolia (Meisn.) Mez against 10 serotypes Salmonella enterica of poultry origin of the highest incidence in the western region of Paraná, as well as to perform the phytochemical screening of the compounds of the two species. After obtaining the aqueous extract and the organic extracts (ethyl alcohol, ethyl acetate and hexane) was determined Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) by technique microdilution broth. Phytochemical prospecting performed according to qualitative methodology of precipitation and colorimetry. The extracts presented inhibitory and bactericidal activity in different concentrations (200 to 25 mg.mL-1) against the serotypes of Salmonella, varying according to the solvent extractor. The ethyl acetate extract presented the best results, followed by the alcoholic, aqueous and hexane extract. It is noteworthy that studies of antimicrobial activity with vegetable extract within the genus Ocotea are scarce, however, the Lauraceae family presents significant biological activities, mainly related to its essential oils. Phytochemical prospecting detected the presence of steroids and alkaloids in both Ocotea species, which may justify the antimicrobial potential found for Salmonella serotypes. It is concluded that these plants represent an alternative for the control of Salmonella in the poultry sector, reflecting a new perspective for studies with natural products.

RESUMO Objetivou-se com este estudo identificar e quantificar os constituintes do óleo essencial de Zanthoxylum caribaeum e avaliar o potencial antimicrobiano de diferentes extratos vegetais e do óleo essencial desta planta frente a diferentes sorotipos de Salmonella enterica, de maior ocorrência e isoladas na região oeste do Paraná, Brasil. A extração do óleo essencial foi realizada utilizando aparelho Clevenger e a composição química foi determinada por Cromatografia Gasosa acoplada a Espectrometria de Massas (CG-EM). Os extratos foram obtidos através de diferentes solventes (etanol, metanol, hexano, acetona, diclorometano, acetato de etila e água destilada). A atividade antibacteriana foi realizada segundo a técnica de microdiluição em caldo. As análises de CG-EM resultaram na identificação de 15 constituintes principais, todos terpenos, representando 63,88 % do total de óleo essencial. Os compostos majoritários identificados foram Germacreno-D (20,77 %), a-Panasinseno (14,40 %) e b-Selineno (11,68 %). O óleo essencial apresentou atividade antibacteriana frente a 5 sorotipos de Salmonella enterica, com CIM e CBM variando de 7000 a 437 µg.mL-1. Já os extratos variam a CIM e CBM de 200 a 25 mg.mL-1, sendo eficazes para a maioria dos sorotipos de S. enterica, com exceção do extrato aquoso. Os resultados sugerem que o óleo essencial e os extratos das folhas de Z. caribaeum representam uma alternativa para o controle de S. enterica, no setor avícola, refletindo uma nova perspectiva para estudos com produtos naturais.

SUMMARY The present study aimed to identify and quantify the components of the essential oil of Zanthoxylum caribaeum and to evaluate the antimicrobial potential of different plant extracts and the essential oil of this plant against different serotypes of Salmonella enterica, which is of greater occurrence and isolated in the western region of Paraná, Brazil. The extraction of the essential oil was performed using Clevenger apparatus and the chemical composition was determined by Gas Chromatography coupled to Mass Spectrometry (GC-MS). The extracts were obtained through different solvents (ethanol, methanol, hexane, acetone, dichloromethane, ethyl acetate and distilled water). The antibacterial activity was carried out according to the broth microdilution technique. The GC-MS analysis resulted in the identification of 15 main components, all terpenes, representing 63.88% of the total essential oil. The major compounds identified were Germacrene-D (20.77%), a-Panasinsene (14.40 %) and b-Selinene (11.68 %). The essential oil presented antibacterial activity against 5 serotypes of Salmonella enterica, with MIC and MBC ranging from 7000 to 437 μg.mL-1. While MIC and MBC from the range from 200 to 25 mg.mL-1, being effective to most serotypes of S. enterica, with the exception of the aqueous extract. The results suggest that the essential oil and leaf extracts of Z. caribaeum represent an alternative for the control of S. enterica in the poultry sector, in this way reflecting a new perspective for studies with natural products.

RESUMO O objetivo com este trabalho foi avaliar in vitro a eficácia de desinfetantes comumente utilizados e biodegradáveis na sanitização de aviários frente a sorotipos de Salmonella spp. isolados de granjas de aves no Estado do Paraná, Brasil.Os experimentos foram realizados utilizando o tempo de contato de 20 minutos e os desinfetantes testados emtrês concentrações:a recomendada pelo fabricante,metade da concentração recomendada e o dobro da mesma,bem como na presença e ausência de matéria orgânica. Em geral, todos os desinfetantes foram capazes de causar a redução significativa nas contagens dos micro-organismos em relação ao grupo controle. Na ausência de matéria orgânica, os desinfetantes a base de ácidos orgânicos apresentaram os melhores resultados frente ao sorotipo S. Gallinarum biovar Gallinarum, e os desinfetantes a base de glutaraldeído e hipoclorito de sódio frente aos sorotipos S. Typhimurium e S. Intantis. Na presença de matéria orgânica, como comumente se encontra o ambiente avícola, pôde-se observar que todos os desinfetantes apresentaram sua eficácia diminuída, sendo o a base de ácidos orgânicos o que apresentou as menores contagens de unidades formadoras de colônias (UFC). O desinfetante a base de ácidos orgânicos mostrou-se uma potencial alternativa de uso na avicultura, pela eficácia no controle dos sorotipos de Salmonella isoladas de granjas avícolas.

SUMMARY The objective of this study was to evaluate the efficacy in vitro of disinfectants commonly used in sanitizing poultry farms and biodegradable disinfectants against Salmonella serotypes isolated from poultry farms in the state of Parana, Brazil. The experiments were performed using the contact time of 20 minutes and disinfectants tested in three concentrations recommended by the manufacturer, half the recommended concentration and twice the recommended concentration, as well as the presence and absence of organic matter. In general, all disinfectants caused significant reduction in the microorganisms counts compared to the control. In the absence of organic matter, disinfectants based on organic acids showed the best results against the serotype Gallinarum biovar Gallinarum and disinfectants based onglutaraldehyde and sodium hypochlorite against the serotypes Typhimurium and Intantis. In the presence of organic matter, as commonly found in the poultry environment, it was observed that all disinfectants showed decreased effectiveness, and the disinfectant based on organic acids had the lowest CFU counts. Accordingly, the disinfectant based on organic acids proved to be potential alternative for use in poultry due to effectiveness in controlling Salmonella serotypes isolated from poultry.

O presente estudo foi conduzido para verificar o perfil de resistência de diferentes sorotipos de Salmonella spp. isolados em aviários de frango de corte frente a agentes antimicrobianos. Foram processados 342 suabes de arrasto provenientes de granjas avícolas do oeste do Paraná, no período de janeiro de 2010 a janeiro de 2011, sendo isoladas 39 amostras de Salmonella spp. Os sorotipos mais frequentes foram: S. Heidelberg, S. Mbandaka, S. Newport, S. Schwarzengrund, S. Enteritidis, S. Livingstone, S. Orion, S. Give e S. Infantis. A determinação do perfil de resistência para os 19 sorotipos de Salmonella identificados foi realizada em relação a 12 antimicrobianos comerciais. Os resultados indicam que 51% dos sorotipos de Salmonella apresentaram resistência a um ou mais antimicrobianos, com 12 diferentes padrões de resistência. O maior percentual de resistência foi verificado à tetraciclina (30,8%), e o menor à gentamicina e cloranfenicol (2,6%). Os níveis de resistência indicam que os antimicrobianos devem ser utilizados nos aviários de forma mais prudente, buscando, assim, minimizar a disseminação de cepas resistentes.

Te present study was carried to verify the resistance profile of different Salmonella spp. serotypes isolated from a poultry broiler house against antimicrobial agents. Tree-hundred and forty two drag swabs from poultry farms in western Paraná were processed in the period from January 2010 to January 2011, and 39 Salmonella spp. Strains were isolated. The serovars were mostly: S. Heidelberg, S. Mbandaka, S. Newport, S. Schwarzengrund, S. Enteritidis, S. Livingstone, S. Orion, S. Give and S. Infantis. The determination of the resistance profile to the 19 identified Salmonella serotypes was evaluated against 12 antimicrobial commercials. The results indicate that 51% of Salmonella serotypes showed resistance to on or more antimicrobials, with 12 different resistance patterns. The highest percentage of resistance was related to tetracycline (30,8%) and the lowest one to gentamicin and chloramphenicol (2,6%). The resistance levels indicate that antimicrobials should be used on poultry farms more carefuly, thus seeking to minimize the spread of resistant strains.

Vários estudos estão sendo conduzidos para a descoberta de novos agentes antimicrobianos provenientes de plantas, para que possam ser utilizados em produtos farmacêuticos, cosméticos e na indústria alimentícia. A ausência de padronização de métodos utilizados para a avaliação de extratos vegetais com potencial antimicrobiano dificulta a comparação de resultados. Considerando a necessidade de estabelecer um método com resultados consistentes para avaliar a atividade antimicrobiana dos extratos vegetais, este trabalho propôs-se a avaliar a atividade antimicrobiana e determinar a concentração mínima inibitória de extratos de plantas da família Myrtaceae sobre diferentes micro-organismos, utilizando três métodos para avaliação de antimicrobianos. Os métodos empregados foram microdiluição em caldo e difusão em ágar por disco e poço. Foram avaliados os extratos de Psidium guajava, Myrciaria cauliflora e Syzygium cumini sobre bactérias Gram-positivas, negativas e levedura. Em geral, a inibição promovida pelos extratos no teste de difusão em ágar por poço foi maior do que os valores obtidos por disco, independentemente do extrato vegetal testado. Contudo, a atividade inibitória de todos os micro-organismos só pôde ser determinada com o método de microdiluição em caldo, que também apresentou os resultados mais reprodutíveis, e mostrou-se o mais econômico e confiável para se avaliar a atividade antimicrobiana de extratos vegetais quando comparado aos outros métodos

Several studies have been conducted to discover new antimicrobial agents from plants to be used in pharmaceuticals, cosmetics and in the food industry. The lack of standardized methods for the evaluation of plant extracts with antimicrobial potential complicates the comparison of results. Taking into consideration the need to establish a method with consistent results to evaluate the antimicrobial activity of plant extracts, this study aimed to evaluate the antimicrobial activity of extracts of some plants from the Myrtaceae family on different micro-organisms based on a comparative assay with three methods that are commonly used for the assess antimicrobials. The methods used were broth microdilution, and agar diffusion by disc and well. The evaluated extracts were those of Psidium guajava, Myrciaria cauliflora, and Syzygium cumini for Gram-positive and negative bacteria and yeast. In general, inhibition extracts promoted by the agar diffusion test by well was higher than the values obtained by disc, regardless of the plant extracts tested. However, the inhibitory activity of all micro-organisms was only possible to be determined with the microdilution broth method, which also presented the more reproducible results, and proved to be the most economic and reliable way to evaluate the antimicrobial activity of plant extracts in contrast with other methods

A fabricação de linguiça do tipo frescal requer uma série de etapas de manipulação, o que eleva as possibilidades de contaminação por diversas espécies de micro-organismos patogênicos ou deterioradores. Isso pode comprometer a qualidade microbiológica do produto final e veicular enfermidades a partir de seu consumo. No período de abril a setembro de 2009, foi avaliada a qualidade microbiológica de 40 amostras de linguiça tipo frescal produzidas artesanalmente e inspecionadas pelo Serviço de Inspeção Estadual e Federal nos municípios de Cascavel e Toledo (PR), através da quantificação de coliformes termotolerantes, contagem de Staphylococcus spp., bactérias mesófilas aeróbias estritas e facultativas viáveis, pesquisa de Salmonellaspp., Campylobacter spp., Bacillus cereus, Clostridium sulfito-redutores e bolores e leveduras. Os resultados revelaram que 55% das amostras analisadas de linguiça tipo frescal inspecionadas e artesanais apresentaram-se fora dos parâmetros estabelecidos pela Resolução RDC nº 12, de 2 de janeiro de 2001, da Agência Nacional de Vigilância Sanitária (Anvisa), em pelo menos um dos grupos de micro-organismos estudados. Portanto, pode-se dizer que a linguiça tipo frescal comercializada no oeste do Paraná pode oferecer riscos à saúde da população.

The manufacture of fresh pork sausage requires a series of manipulation steps, which increases the chances of contamination by several species of micro-organisms, be them pathogenic or deteriorating. This may compromise the microbiological quality of the final product and transmit diseases with consumption. In the period from April to September 2009, the microbiological quality of 40 samples of fresh handmade sausage inspected by the Inspection State and Federal Service in the cities of Cascavel and Toledo (PR) was assessed, through the quantification of thermotolerant coliforms, Staphylococcus spp., viable facultative and strict mesophilic aerobic bacteria, Salmonella spp., Campylobacterspp., Bacillus cereus, Clostridium sulfite reducers, molds and yeasts. The results revealed that 55% of the analyzed samples of fresh handmade and inspected sausages were not in accordance with the parameters established by Resolution RDC Nº 12, of January 2nd, 2001, from ANVISA, in at least one of the studied groups of micro-organisms. Therefore, we can say the sausages sold in western regions, may present a risk to public health.

Extracellular proteases have been shown to be factors of virulence in fungal entomopathogenicity. We examined the production of the cuticle-degrading extracellular proteases chymoelastase (Pr1) and trypsin (Pr2) in isolates of the fungus Metarhizium flavoviride. Fungal growth was in a mineral medium (MM) containing nitrate, and in MM supplemented with either cuticle from Rhammatocerus schistocercoides or with the non-cuticular substrate casein. The substrates used for growth influenced the expression of both analyzed proteases, the highest protease activities of nearly all isolates having been observed in the medium containing insect cuticle, with more Pr1 than Pr2 being produced. There was a natural variability in the production of cuticle-degrading proteases among isolates, although this was less evident for Pr2. Our data support the hypothesis that the production of Pr1 on insect cuticle is a useful characteristic for the analysis of intraspecific variability of M. flavoviride isolates.
